Oxidative stress results from an imbalance between ROS production and the intracellular and extracellular antioxidants that scavenge ROS. There are many direct assays that measure the oxidation of the sperm cell membrane. The most used assay measures malondialdehyde (MDA), one of the final products of sperm cell membrane lipid peroxidation. Increased levels of MDA correlate with decreased sperm parameters.
Quantification of sperm DNA damage has also been used as assay for intracellular ROS-induced oxidant injury by measuring a specific product of oxidant-induced DNA damage, 8-oxo-7, 8, -dihydro 2′ deoxyguanoside (8-OHdG), used as a specific marker of oxidative injury to sperm DNA .
